If I receive a rude, offensive letter full of lies and innuendo that's merely a direct, uninterrupted communication from them to me, could that be considered defamation of character?

I believe that this can really be counted as a defamation of character and in this case it is worth immediately contacting a specialist who knows how to prove defamation damages fairly and without unnecessary problems. As the saying goes: modern problems require a modern solution, right? that is why I advise you not to waste time and stop suffering such attacks in your address and deal with it once and for all. With professional advice, you can get out of this and get rid of such emails
